Considered the youngest of all the Ham-Hams, Penelope is usually never seen without the cover of her yellow blanket (though a few seconds in the opening theme show her blanket coming off, revealing her brown fur). Although she is friendly to everyone, she feels particularly attached to Pashmina. Being innocent and naive, due to her age, she gets into trouble more often than most of the other Ham-Hams.   In season two, she has shown some cross-species affection for Herbert the Pig, a boy by the name of Ethan, and a rabbit at Ethan's school. Unlike the other Ham-Hams, and perhaps because of her young age, Penelope can't say anything more than 'Ookwee!' or 'Ookyoo!' ('Ukyu!' in the original version).

A member of S.T.A.R.S.(Special Tactics And Rescue Service) assigned to Alpha Team under Captain Albert Wesker. During the mansion incident, they discovered that the pharmaceutical company called Umbrella Corporation created a mutagenic virus known as the T-Virus, along with other experiments. Jill, along with Chris, Barry, Rebecca and Brad survived the mansion incident and upon returning to Raccoon City their story was met with skepticism prompting the other three S.T.A.R.S members to head off on their own investigations, while Jill and Brad remained in Raccoon City to investigate an underground research facility of Umbrella. On September 28, 1998, the city became infected by the T-Virus, the Umbrella corporation deploys the Nemesis-T Type was deployed to hunt down the remaining S.T.A.R.S members, Jill survives the ordeal along with a U.B.C.S operative named Carlos Oliviera when the US Government decided to eradicate the city using an experimental thermobaric missile. After the fall of Umbrella, she along with Chris Redfield and Barry Burton went on to found the BSAA (Bioterrorism Security Assessment Alliance). She joins Chris Redfield in a number of missions namely in pursuit of II Veltro, the raid of the Spencer Estate wherein she was presumed to have died and the Kijuju incident where she, under the control of Albert Wesker, would end up fighting Chris Redfield and his partner Sheva Alomar. Chris successfully saves Jill from Wesker's control and by the end she assists Chris and Sheva in bringing down Wesker.

Jill Valentine would star in various non-canon appearances such as the Marvel vs Capcom franchise, SNK vs Capcom, We Love Golf!, and Project X Zone. She was portrayed by Sienna Guillory in the live action Resident Evil films, and will be played by Hannah John-Kamen in the upcoming Resident Evil reboot film.
