The film centers on the sextuplets at a high school class reunion. There, they see all their former classmates who have become "proper" adults with company jobs. The sextuplets sulk about this and go home and drink until they fall asleep. But when they wake up, a certain scene awaits them.

The Matsuno sextuplets (leader Osomatsu, wannabe tough guy Karamatsu, straight man Choromatsu, cynical Ichimatsu, cheerfully stupid Jyushimatsu and lovable spoiled Todomatsu) are now in their 20s, still living together in the same house as NEETs. Their neighbors still include such eccentric characters as fish market idol Totoko, loud bucktoothed troublemaker Iyami, tiny rival Chiba, wacky inventor Dekapan, and giant mouthed glutton Dayon. 
 
Join the Matsuno siblings as they overcome mundane hurdles in their daily lives, such as finding employment, figure out who should pay the bar bill, and who should get the extra pastry!

Matsuno household has six naughty and mischievous sons (who are sextuplets), led by the eldest Osomatsu. They were grade schoolers back in the Showa period, but despite of still living in the same old household, the street view as well as the life style of modern day society have changed drastically.
 
The manga holds an original story, completely different from the one in the anime.

Prologue shorts included with the BD/DVD release of Mr. Osomatsu The Movie. A single short was screened in theaters per how many weeks the movie was playing, with each acting as a prologue to the movie.
